Abstract
Lipid in the epidermis has unique features and plays a key role in skin barrier. This lipid barrier prevents the loss of fluid and electrolytes from the internal milieu, while simultaneously preventing the hostile external environment. Lamellar bodies play an essential role in delivering the lipids into the extracellular spaces of the stratum corneum. The extracellular lipid matrix is composed of predominantly ceramides, cholesterol and free fatty acids, organized into structurally unique lamellar bilayers. Alterations in lipid content and composition result in impaired barrier function and are associated with a number of skin diseases such as atopic dermatitis. This review describes ceramides, cholesterol, and fatty acids synthesis in the epidermis, and their important role in the formation and maintenance of the skin barrier.
